Acute Effects of Exercise on Breast Cancer Biomarkers: The ACE Study

An active lifestyle improves outcomes for women with breast cancer. The ACE Study, funded by the Breast Cancer Research Foundation, will investigate how a 45-minute bout of moderate exercise can affect the biology of the human body. This could help explain the influence of physical activity in women with breast cancer.You may be eligible to join the ACE study if you are:– aged 36-75– no menstrual period for the past 12 months– have been diagnosed with breast cancer Stage 0-3c (i.e., breast cancer that hasn’t spread to other parts of the body) within the past 5 years– finished your main treatment at least 6 months ago, but can still be on long term hormone treatment– can visit our exercise center for two clinic visits at the Fred Hutch Cancer Center’s Prevention Center at South Lake Union, Seattle WA.If you are eligible and consent to participate, we will invite you to a clinic visit at the Fred Hutch for a physical examination to ensure that it is safe for you to exercise. If you are eligible based on your test results, and would still like to participate, a computer program will assign you by chance to one of the following groups:1. Exercise Group: Participants in this group will have a 45-minute bout of exercise on a stationary bike2. Resting Control Group. Participants will sit for 45 minutes.At a second visit, you will be asked to give a blood sample, a sample of fat tissue from your abdomen, and a sample of muscle tissue from your thigh. These samples will be collected under sterile conditions using a needle and will be about the size of a pencil eraser. Then, you will either exercise or rest for 45 minutes, and give another blood sample, and samples of fat and muscle tissue. A third blood sample will be collected one hour later.Benefits to Participants.– Participants will receive information about their study test results, and a brochure that will provide individuals with tips and advice to help achieve success with their current of potential fitness plan– At the end of the second visit, all study participants who give blood and tissue samples will receive $200.More details including a link to a screening questionnaire can be found at https://theacestudy.org/

Participant Eligibility

– Female and aged 36–75 years
– A history of Stage 0-3 breast cancer diagnosed within the past 5 years
– Completed treatment at least 6 months ago (if you are taking long-term hormone treatment you may still be eligible)
– Able to travel to the Fred Hutch Cancer Center in South Lake Union, Seattle for 2 clinic visits. Each visit will take between 2-3 hours
– Able to exercise safely at a moderate intensity for 45 minutes
– Willing to be assigned by chance by a computer program to either the exercise or control (45 mins resting period) group
– Consent to release medical records to verify cancer diagnosis and to review health information
– No menstrual period for the past 12 months (due to either menopause, surgery or a side-effect
of treatment)
– Willing to undergo a muscle and adipose tissue biopsy before and after the exercise and control. The amount of tissue biopsied will be about the size of a pencil eraser

You will NOT be eligible if you
– Have a history of diabetes, or are taking medications to treat high blood sugar
– Have a history of cancer other than breast (other than non-melanoma skin cancer)
– Have ever been told by their doctor that you should NOT exercise
– Have a clotting disorder
– Have an allergy to local anesthetics
– Are using any sex hormones (e.g., estrogen, progesterone, testosterone or other androgens, for the previous 3 months) of any type including oral, creams, vaginal creams or inserts, patches, implants. if you are taking long-term anti-estrogen therapy – e.g., tamoxifen, aromatase inhibitors you may still be eligible
– Are using warfarin or NSAIDs on the advice of your doctor, weight loss medications, statins, oral beta-blockers
– Drink more that 2 alcoholic drinks per day
– Use any tobacco products including smoking, vaping, chew, or nicotine patches
– Are a frequent marijuana user (>1 per month)
